I am considering purchasing the My Cloud PR4100. At this point, I can’t really justify buying all four drives, so I was wondering if I would be able to upgrade from an initial 2-drive RAID 1 to a 4-drive RAID 10 at a later time without having to reformat and lose all of my data. Does WD support this?

If you go from RAID 1 to 10, you would have to reformat the drive in order to have the volume created. However, you should be able to create another RAID 1 with the 2 new drives, but switching to RAID 10 is not possible without a reformat.
